The NSM2 as virus effector: targets, topology and functional consequences in T cells

Surface contact with Measles virus (MV) is known to paralyze cytoskeletal dynamics and proliferative responses of T cells. Activation of the neutral sphingomyelinase 2 (NSM2) is centrally involved in this process. In the frame of this project, we will define effectors, signalling pathways and functional consequences of NSM2 activation in MV-induced T cell paralysis by
- identification and functional validation of NSM2 early membrane proximal signalling components and their downstream effector pathways
- identification and functional validation of components of the NSM2-associated proteome essential for its activity, and
- addressing the role of NSM2 compartmentalization in response to MV signalling in T cells.
In addition to basic and advanced virological, cell biological and biochemical methods, the project involves advanced microscopy and also integrates systems biology and bioinformatic processing.
